Background: There is no universally accepted behavioral scoring to define the early development of pentylenetetrazole (PTZ) kindling. Therefore, studies investigating alterations of neurogenesis in the PTZ model were mainly focused on full kindled animals rather than early stages of kindling. This study aimed to determine an appropriate behavioral index for categorizing stages of PTZ kindling progress and to evaluate neurogenesis during PTZ kindling. Materials and Methods: Twenty-four mice were intraperitoneally injected with a sub-convulsive dose of PTZ (40mg/kg) every other day until they became full kindled. The first occurrence of different seizure behaviors and their durations were recorded during kindling development, and the different stages of kindling were categorized. Neurogenesis was evaluated in the lateral subventricular zone (SVZ) at each stage of kindling by immunofluorescence staining. Results: First occurrence of restlessness, motionless staring, hind limb tonic extension, Straub’ s tail, myoclonic jerk, and tonic-clonic were sequentially observed in more than 80% of animals with increasing PTZ injections. The duration of the myoclonic jerk was significantly longer than the other seizure behaviors. The significantly higher percentage of BrdU-positive cells was found in SVZ of mice showing tonic-clonic in comparison to other seizure behaviors. Conclusion: A hierarchy behavior was observed during the kindling process when considering the first occurrence of seizure behaviors. We defined the first occurrence of restlessness, motionless, hind limb tonic extension and Straub’ s tail behaviors as an early phase, myoclonic jerk as a borderline phase and tonicclonic as a late phase of PTZ-induced kindling. Our results indicated an enhanced SVZ neurogenesis at the late phase of kindling.

Vitamin A and its derivatives have recently reported to be implicated in synaptic plasticity. The possible effect of vitamin A and its precursor, b-carotene, on seizure acquisition was tested in PTZ kindling model of epilepsy. Vitamin A and b-carotene were tested for their ability to 1) suppress seizures (clonic and tonic) and lethality induced by PTZ in PTZ-kindled mice (anticonvulsant effect) and 2) attenuate the development of sensitization to convulsive and lethal effects of PTZ in kindled mice (anti-epileptogenic effect) when given as a pretreatment prior to each PTZ injection during kindling acquisition. Diazepam was used as positive control group. All these drugs showed anti-epileptogenic effects against PTZ-induced tonic seizures and lethality. Vitamin A and b-carotene had no effect on clonic seizures induced by PTZ in kindled mice. It can be concluded that vitamin A and b-carotene possess anti-epileptogenic activity. Retinoids appear to be valuable for development of a new class of anticonvulsant drugs.

مقدمه: با توجه به اثرات متقابل مغز و طحال، در این پژوهش اثر اسپلنکتومی بر صرع ناشی از پنتیلن تترازول در موش صحرایی نژاد ویستار مورد بررسی قرار گرفته است.مواد و روش ها: در این پژوهش از 48 سر موش صحرایی نر و ماده نژاد ویستار، در 3 گروه استفاده شد. گروه های آزمایشی برای هر یک از دو جنس شامل شاهد منفی، شاهد مثبت و اسپلنکتومی بود. گروه شاهد منفی تحت عمل جراحی قرار نگرفتند. در گروه شاهد مثبت برش جراحی ایجاد گردید، ولی طحال خارج نشد. در گروه اسپلنکتومی طحال خارج شد. پس از 7 روز، پنتیلن تترازول تزریق و زمان شروع مراحل مختلف تشنجی برحسب ثانیه و عمق حملات تشنجی در 3 گروه به تفکیک جنس، با هم با استفاده از آزمون آنالیز واریانس یک طرفه مقایسه شد.نتایج: زمان شروع مرحله دوم تشنج در گروه اسپلنکتومی موش های نر در مقایسه با گروه های شاهد، کاهش معناداری نشان داد (P<0.05). زمان شروع مرحله چهارم تشنجی در گروه اسپلنکتومی از موش های ماده نسبت به گروه شاهد منفی مربوط به این موش ها، کاهش معناداری داشت. همچنین از لحاظ امتیاز تشنج در 2 گروه موش های نر و ماده، در هر 3 گروه تفاوت معناداری مشاهده نشد.نتیجه گیری: اسپلنکتومی موجب تشدید حملات تشنجی ناشی از پنتیلن تترازول در موش صحرایی نژاد ویستار می شود.

Introduction: Guaifenesin is an expectorant used commonly as an OTC medication. It is also used as a muscle relaxant in anesthetic protocols in veterinary medicine. There have been some reports that guaifensin has beneficial effect in fibromyalgia due to its possible NMDA antagonist activity. Considering that drugs with similar structure to guaifensin (i.e. flebamate) and those with NMDA antagonist activity or muscle relaxant properties have been clinically used as an anticonvulsant; the aim of this study was to determine whether guaifensine has an anticonvulsant effect in an animal model of seizure.Materials and methods: Anticonvulsant effect of guaifensin was assessed by pentylenetetrazole (PTZ)-induced convulsion. Male albino mice were received guaifensine (100, 200, 300 or 400 mg/kg; n=8-10), 0.25% tween (vehicle) or Diazepam (3mg/kg, n=8; reference drug) intraperitonealy 30 minutes before the injection of PTZ (95mg/kg). The latency time before the onset of myoclonic, clonic and tonic-clonic convulsions, the percentage of animals shown convulsion and the percentage of mortality were recorded. In addition, the effect of guaifensin on neuromuscular coordination was assessed using rotarod.Results: Guaifensin at doses higher than 200 mg/kg significantly increased the latency of myoclonic, clonic and tonic-clonic convulsions in a dose-dependent manner. ED50 for protection against PTZ-induced clonic, tonic-clonic seizures and death were 744.88, 256 and 328 mg/kg, respectively. Guaifensin at doses higher than 100 mg/kg significantly reduced neuromuscular coordination compared to vehicle-treated group.Conclusion: These results suggest that guaifensin have muscle relaxant and anticonvulsant effects and may have a potential use in convulsion.
